Validasi Spektrofotometer UV-VIS pada Analisis Formalin Di Poltekkes Kemenkes Pontianak

Abstract


Abstract: Validation is the process of determine a method, an instrument and a technician to fulfl the requirements for testing. Validation of the instrument needs to be done separately. The UV-VIS spectrophotometer in Poltekkes Kemenkes Pontianak has many advantages such as wave length 190 - 1100 nm, using double beam optical system, has 2 nm spectrum bandwidth, but the using of the instrument has not been maximized. It is necessary to validate the UV-VIS spectrophotometer. The aim of this research is to determine the precision, accuracy, linearity, Limit of Detection (LOD) and Limit of Quantifcation (LOQ) on UV-VIS spectrofotometer on formalin analysis at Poltekkes Kemenkes Pontianak. The research uses pre-experimental design and purposive sampling method with formalin standard as population. The sample used is formalin standard with various concentration 0 ppm, 1 ppm, 2 ppm, 3 ppm, 4 ppm and 5 ppm with nine replication. Samples reacted with chromatophore acid, the purple color showed the sample containing formalin, then absorbance value measured to obtain formalin content in the sample. The result of precision value 94.43%, accuracy value 97.77%, linearity test in the concentration range 0-10 ppm gives correlation coeffcient value r = 0.918 with the limit of detection (LOD) of the formalin is 4.98 ppm and the limit of quantifcation (LOQ) of the formalin is 16.60 ppm. Based on the result of the research that it can be concluded that UV-VIS spectrophotometer on formalin analysis in Poltekkes Kemenkes Pontianak well validated because of the fulfllment of fve validation parameter criteria.

Abstrak: Validasi adalah proses untuk memastikan apakah suatu metode, alat dan teknisi memenuhi persyaratan untuk pengujian. Validasi alat perlu dilakukan tersendiri. Spektrofotometer UV-VIS yang ada di Poltekkes Kemenkes Pontianak memiliki banyak kelebihan diantaranya rentang gelombang yang lebih lebar 190 – 1100 nm, menggunakan sistem optik double beam, memiliki bandwidth spektrum 2 nm, namun penggunaannya belum maksimal. Maka perlu dilakukan validasi terhadap spektrofotometer UV-VIS tersebut. Tujuan penelitian ini untuk mengetahui presisi, akurasi, linearitas, batas deteksi (LOD) dan batas kuantitasi (LOQ) pada Spektrofotometer uv-vis pada analisis formalin di Poltekkes Kemenkes Pontianak. Penelitian ini menggunakan desain pre-experimental metode purposive sampling dengan populasi standar formalin. Sampel yang digunakan yaitu standar formalin dengan berbagai konsentrasi 0 ppm , 1 ppm, 2 ppm, 3 ppm, 4 ppm dan 5 ppm dengan pengulangan sebanyak sembilan kali. Sampel direaksikan dengan asam kromatofat, perubahan warna menjadi ungu menunjukkan sampel mengandung formalin, kemudian diukur nilai absorbansinya untuk mendapatkan kadar formalin dalam sampel. Hasil penelitian nilai presisi 94.43%, nilai akurasi 97.77%, uji linearitas pada rentang konsentrasi
0 – 10 ppm memberikan nilai koefsien korelasi r = 0.918 dengan batas deteksi (LOD) formalin 4.98 ppm dan batas kuantitasi (LOQ) 16.60 ppm. Berdasarkan hasil penelitian dapat disimpulkan bahwa spektrofotometer UV-VIS pada analisis formalin di Poltekkes Kemenkes Pontianak tervalidasi dengan baik karena terpenuhinya kriteria lima parameter validasi.
